چکیده

Stimulated emission and lasing have been demonstrated for chlorophyll a, chlorophyll b, and bacteriochlorophyll a as well as for the Mg-free chlorophyll derivatives pheophytin a and methyl pheophorbide a. Population inversion was obtained by optically pumping with an N(2) laser. For the three chlorophylls, lasing was observed for only the (0,0) vibronic region of the S(1) --> S(0) fluorescence band. In contrast, lasing was observed for both the (0,0) and (0,1) vibronic regions of the S(1) --> S(0) fluorescence band of pheophytin a and methyl pheophorbide a. Differences between the lasing behavior of the chlorophylls and the Mg-free derivatives probably reflect differences in aggregation behavior. Laser pulses of duration shorter than the fluorescence lifetimes were generated. This result suggests that stimulated emission must be considered in the interpretation of fluorescence lifetime data for in vitro and in vivo chlorophyll-containing systems.

Photometric method for the quantification of chlorophylls and their derivatives in complex mixtures: fitting with Gauss-peak spectra.

Accurate quantification of pigments in mixtures is essential in all cases in which separation of pigments by chromatography is impracticable for one reason or another. An example is the analysis of in vivo formation of heavy metal-substituted chlorophylls in heavy metal-stressed plants. We describe here a novel, accurate UV/VIS spectrophotometric method for the quantification of individual chlo...

In vitro Conversion of Vinyl to Formyl Groups in Naturally Occurring Chlorophylls

The chemical structural differences distinguishing chlorophylls in oxygenic photosynthetic organisms are either formyl substitution (chlorophyll b, d, and f) or the degree of unsaturation (8-vinyl chlorophyll a and b) of a side chain of the macrocycle compared with chlorophyll a. We conducted an investigation of the conversion of vinyl to formyl groups among naturally occurring chlorophylls. We...

Root-shoot interaction in the greening of wheat seedlings grown under red light.

Wheat seedlings grown with roots exposed to constant red light (300-500 micromoles m-2 s-1) did not accumulate chlorophyll in the leaves. In contrast, seedlings grown with their roots shielded from light accumulated chlorophylls. Chlorophyll biosynthesis could be induced in red-light-grown chlorophyll-deficient yellow plants by either reducing the red-light intensity at the root surface to 100 ...

Haems and chlorophylls: comparison of function and formation.

In 1844 Verdeill reported that acid treatment of chlorophyll or haem yielded apparently similar red compounds; he even postulated that chlorophylls would contain iron. Hoppe-Seyler2 confirmed the apparent similarity of acid derivatives of haems and chlorophylls from their light absorption characteristics, a point rather overshadowing the discovery at the same time by McMunn3 of cytochromes, ano...

Light-Induced Accumulation and Stability of Chlorophylls and Chlorophyll-Proteins during Chloroplast Development in Radish Seedlings

Illumination of 3 day old etiolated radish seedlings with continuous white light results in a progressive accumulation of chlorophyll a and b. Both pigments are bound in a different way to the thylakoid chlorophyll-proteins, which appear parallel to the formation of chlorophylls. By applying the SDS-PAGE method to SDS-digested chloroplasts, it was possible to show that the chloroplasts of radis...
